To understand the genetic basis of human Type 1 diabetes (T1D), I am using mouse models to define the mechanism specific genes play in disease susceptibility and resistance. Previously we have identified a gene, encoded within the mitochondrial genome, that is protective against T1D in both humans and mice. This gene changes mitochondrial function. Mitochondria are essential for the life and death of cells, and free radicals produced by these organelles modulate cellular functions. One of my major goals is to understand how mitochondria affect two important cellular players in T1D, T cells and beta cells. My second goal is to elucidate the interactions occurring between the nuclear and mitochondrial genomes. The cross talk between these two genetic compartments is likely essential for controlling free radical production and cellular function of human beta cells as well as activation of immune cells.
